Preparation of microscope slide strains for yeast detection
Place a round piece of filter paper at the bottom of the petri dish plastic disposable and place a U-shaped glass rod on top of it with two clean microscope slides lab. After sterilization, a few milliliters of sterilized 20% peroxide solution was added to the filter paper in a sterile way. The main function of the solution was to keep the filter paper moist during the culture process. Using a disposable pasteur pipettes, melted yeast extract AGAR or potato glucose AGAR is added to the surface of the slide to form a very thin AGAR culture substrate. The yeast to be tested was inoculated on the microscope slides laboratory by inoculation ring line, part of the medium was covered with a sterile microscope glass cover, and the sterile disposable petri dish was covered with a cover and cultured at 25℃ for more than 7d.
The phase contrast microscope was used to observe the growth condition of the cells, and the growth morphology of the cells exposed to air and under the cover glass was compared. The cover glass can also be gently removed from the slide, drop a drop of milk phenol, milk phenol-picric acid or milk phenol cotton blue to fix the bacteria, and then cover the cover glass, the edge of paraffin wax, nail polish or special adhesive seal.
1. Wipe the glass slides microscope laboratory and cover glass microscope.
2. Place a drop of water in the center of the medical microscope slides.
3. Pick a few hyphae with an dissecting needle.
4. Apply to clear water droplets.
5. Cover with ccover glass microscope slides.
6. If you need to stain, you should stain, and you can observe without staining.
